I create my mug designs in AI as 216x91mm artboards, but struggle to have designs flush (double sided).
Can anyone suggest a way i can have two specific target areas within the artboard o nthe left and right, which will always print out flush? I use BMS Precut paper.

Create your artboard. On the layer place ruler guides on the left hand edge and the centre of the artboard then lock the layer. Make a new layer. Turn off smart guides and untick snap to guides Make your first target area (no fill 1mm stroke black) on the left side and position it correctly on a layer then lock the layer. Make a new layer. draw a rectangle (no fill 1mm stroke black) on this layer and drag it out to cover half the artboard ie from one ruler guide to the other. Now unlock both layers and select the two shapes. Copy them and paste onto a new layer and group them. Lock other layers. Now use the selection tool and drag the left hand centre handle all the way over to the right edge. [or select transform reflect] This should give you two identical (mirrored shapes). Then unlock all layers and delete the outer rectangles. You will now have two rectangles placed in identical positions at each end of the artboard. You can then use the rectangles you have left to place your designs and, when you are happy turn off the rectangle layers to show only your design.
